Recombinant influenza virus carrying bicistronic vRNA encoding two genes arranged in tandem |
abstract |
The present invention relates to a recombinant influenza virus for high-yield expression of an integrated foreign gene, wherein the recombinant influenza virus is genetically stable in the absence of a helper virus and comprises at least one virus. An RNA segment, wherein the viral RNA segment is a tandem bicistronic RNA molecule encoding two tandem genes, in which one of the standard viral genes is covalently linked to an exogenous recombinant gene. And the viral RNA segment has an upstream splice donor and a downstream splice acceptor signal surrounding the closer coding region. Further, the present invention provides a method for obtaining an attenuated virus against recombination-dependent progeny production in the case of opportunistic superinfection with a wild-type influenza virus; a method for producing the recombinant influenza virus; Compositions; and the use of said recombinant influenza virus for the manufacture of a medicament for inoculation purposes. |
