The present invention provides cells capable of expressing glucocerebrosidase (GCB) under conditions that prevent removal of at least one mannose residue distal from the pentasaccharide core of the precursor oligosaccharide of GCB. Wherein a method for producing high mannose glucocerebrosidase (hmGCB) comprising producing GCB with a precursor oligosaccharide, thereby producing an hmGCB formulation. Preferably, the condition that prevents removal of at least one mannose residue distal from the pentasaccharide core is inhibition of a class 1 processing mannosidase and / or a class 2 processing mannosidase. The invention also features hmGCB formulations and methods of using hmGCB formulations. |
