titleOfInvention | Methods for identifying compounds that regulate the intracellular distribution of phosphodiesterase (PDE) enzymes in living cells |
abstract | Disclosed are alternative therapies for PDE4 inhibition. Transposers of PDE4 remove PDE4 from its natural location in the cell, thereby increasing cAMP concentration at this location. By translocating PDE4 and thereby not directly acting on the well-conserved site of the catalyst of the phosphodiesterase inhibitor, the compound acts, for example, on the binding domain of PDE4 and thereby the PDE4 isoform-specific " An inhibitor. Transposition of PDE4 is visualized using fusion to GFP. The native position is derived by treatment with rolipram. |
