abstract | The present invention provides a method for quantitatively measuring the amount of nucleic acid generated in association with a polymerase reaction using pyrophosphate generated in association with the polymerase reaction. (1) A target nucleic acid fragment having a known base sequence, at least one primer complementary to a part of the target nucleic acid fragment, at least one deoxynucleoside triphosphate, and at least one polymerase. And a step of performing a polymerase extension reaction starting from the 3 ′ end of the primer using the target nucleic acid fragment as a template, using a reaction mixture containing a divalent cation necessary for the polymerase reaction; and (2) A method of detecting whether or not the polymerase extension reaction proceeds continuously by detecting pyrophosphate generated in association with the polymerase extension reaction; Polymerer is the first reaction to detect the accompanying pyrophosphate The detection method described above, wherein the detection method is carried out simultaneously with a ZE extension reaction.
