abstract An object of the present invention is to provide a novel measurement method for measuring the dissolution or unwinding activity of a double-stranded helical structure of a nucleic acid, or the ability of a single-stranded nucleic acid-specific binding protein. In addition, the present invention provides a simple and highly quantitative and reproducible measuring kit to which the above-mentioned measuring method is applied. The measurement method of the present invention comprises: (1) a first single-stranded nucleic acid probe having a fluorescent energy donor at a 5 ′ end or a 3 ′ end, and a first single-stranded nucleic acid probe having a complementarity with the first probe; A second single-stranded nucleic acid probe having an energy acceptor at the opposite end of one probe is prepared, and (2) fluorescence resonance energy transfer (FRET) is caused by using the pair of nucleic acid probes as a double-stranded nucleic acid. (3) measuring the change in the fluorescence spectrum due to the release of FRET accompanying the release of the double-stranded structure of the double-stranded nucleic acid.
