PROBLEM TO BE SOLVED: To provide a modified human factor VIII cDNA. SOLUTION: This cDNA is a modified human factor VIII cDNA, and has a mutation in either the wild type or the factor VIII cDNA in which the B domain is partially or completely deleted and replaced with a DNA linker segment. A) One or more codons of the human Factor VIII cDNA that are not identical to the codons corresponding to the same position in the porcine Factor VIII cDNA are inserted by any of the following methods: Codons for the oppositely charged amino acid, if the porcine sequence contains codons for charged amino acids, while the human sequence contains codons for charged amino acids, while the porcine sequence contains codons for charged amino acids. B) one or more charged amino acids found in the factor VIII cDNA of a hemophilia patient Codon, consisting of those is substituted with the codon for amino acid of opposite charge related.
