abstract | An object of the present invention is to provide a two-dimensional protein electrophoresis method for a protein, which can easily and quickly analyze a small amount of a sample on a microchip. The microchip includes a loading channel, a separation channel crossing the loading channel, a sample reservoir is disposed at one end of the loading channel, and an outlet is disposed at the other end of the loading channel. In a microchip in which each of the loading channel and the separation channel is filled with an electrophoresis buffer, (A) subjecting a sample containing a protein having a pH at least 2 different from that of the electrophoresis buffer to the sample reservoir, (B) introducing the sample in the sample reservoir to the separation channel by pressurizing the inside of the loading channel; and (c) loading the sample in the separation channel by applying an electrophoretic electric field to the separation channel. A two-dimensional electrophoresis method for simultaneously separating proteins by molecular weight and isoelectric point, which comprises the step of electrophoresis. [Selection diagram] None |
