abstract | Kind Code: A1 The present invention relates to a method for measuring the activity of a drug metabolizing enzyme which is important in a drug metabolizing reaction in a living body, particularly various kinds of mutant cytochrome P450 enzymes, and a drug metabolizing enzyme of a chemical substance, particularly various mutant cytochromes. The present invention relates to a method for evaluating an activity inhibitory effect on a P450 enzyme, and a composition for the method. In a first reaction, a compound serving as a substrate for a drug-metabolizing enzyme is reacted with a necessary cofactor or the like by a wild-type or various mutant drug-metabolizing enzymes, and a product generated in the second reaction is converted to a product. Competitive measurement by immunochemical assay using specific antibodies enables rapid and simple use of various mutant drug-metabolizing enzyme activities and chemical substances without using isotopes or effective analysis. The ability to evaluate the inhibitory effect on the activity of various mutant drug metabolizing enzymes can be evaluated. In order to realize this measurement method, the prepared composition is efficiently and stably constructed. |
