abstract | (57) [Summary]nThe present invention relates to methods for destabilizing proteins in living cells and their use in developing new assays. In one embodiment, the invention includes the use of a noncleavable multimeric ubiquitin fusion protein to destabilize a target protein, such as a reporter moiety. In one aspect of the method of the invention, the construct also comprises a linker operably linking the reporter moiety to the multimerized ubiquitin fusion protein. In this embodiment, the enzymatic modification of the linker regulates the binding between the reporter protein and the multimerized ubiquitin domain, and changes the stability of the reporter moiety. Next, the level of the reporter moiety in the cell can be used as an indicator of the enzyme activity in the cell. In another aspect, the present invention provides a general method for coordinately regulating the intracellular concentrations of a plurality of target proteins. In one aspect of the method of the present invention, a target protein is operably linked to a ubiquitin fusion protein via a linker that includes a protease cleavage site. Cleavage of the linker with a protease dissociates the target protein from the multimerized ubiquitin construct and increases the stability and concentration of the target protein. |
