abstract | (57) [Summary]nThe present invention relates to reverse transcriptases with increased fidelity (or decreased misintegration rate) and / or terminal deoxynucleotidyl transferase activity. Specifically, the present invention relates to a method for producing such reverse transcriptase by modification or mutation of a specific site in the reverse transcriptase. The invention also relates to a nucleic acid molecule comprising a gene encoding the reverse transcriptase of the invention, a host cell comprising such a nucleic acid molecule, and a method of making this reverse transcriptase using the host cell. The reverse transcriptase of the present invention is particularly suitable for nucleic acid synthesis, sequencing, amplification and cDNA synthesis. |
