titleOfInvention | Method and apparatus for detecting enzymatic activity using molecules that alter electrophoretic mobility |
abstract | (57) Abstract: Use of a fluorescently labeled substrate molecule (114) whose electrophoretic mobility is changed by a chemical reaction such that the activity of a chemical reaction of a molecule in a cell is catalyzed by an enzyme (112). Is measured by Specificity is achieved with a labeled substrate molecule (114) that can only be acted upon by a specific enzyme (112). Thus, the activity of one specific enzyme (112) or a class of enzymes can be measured. The measurement uses the presence of such a substrate molecule (114) in the cell and, at a point of interest, typically exposes the cell (46) to a stimulant (118) that activates a particular enzymatic pathway. It is done after doing. To ensure accuracy, measurements must be made in a timely manner to minimize chemical reactions that occur subsequent to the point of interest. Fast controlled laser lysis is used to obtain the contents of a single cell (46) into which the reporter substrate molecule (114) has been introduced. The cell contents are then subjected to capillary electrophoresis (22), and the enzymatic activity is determined by determining the amount of the substrate molecule (114) by an enzyme-altered substrate molecule (separated by electrophoresis and identified by the presence of a fluorescent label). 128). |
