abstract | (57) [Summary]nPROBLEM TO BE SOLVED: To improve the efficiency of uptake of a chain terminator by a family B archaeal DNA polymerase. For some time, the incorporation efficiency of ddNTPs, and more particularly dye-labeled ddNTPs, has limited the usefulness of this group of DNA polymerases in protocols requiring the incorporation of chain termination factors.nIn the present invention, several new things are used in new combinations. In accordance with the present invention, derivatized ddNTPs that are more efficiently incorporated than the corresponding underivatized ddNTPs are identified. A method is described to identify additional compounds of this type. Such compounds offer significant advantages over previously investigated dye-labeled ddNTPs where uptake was undesirable. |
