abstract | (57) [Summary]nThe present invention relates to a method for identifying a transmembrane domain of a transmembrane protein. The transmembrane protein can be a group I receptor, a group II receptor, and a receptor having multiple transmembrane domains. The method involves the use of a death domain deleted protein that has been modified to replace its transmembrane domain with a candidate nucleic acid sequence. The absence of apoptosis in a host cell expressing a modified death domain deleted protein and a death domain containing protein after exposure to an apoptosis inducing ligand is a candidate nucleic acid encoding the transmembrane domain of a transmembrane receptor Indicates the presence of the sequence. Also provided are vectors and host cells that produce the modified death domain deleted proteins. The present invention further relates to a transmembrane protein containing the transmembrane domain identified by the method of the present invention and the identification of a target molecule for the transmembrane protein. The present invention also includes a method for identifying a substance that alters the interaction between a transmembrane protein identified by the method of the present invention and a target molecule. |
