Assay for caspase activity using green fluorescent protein |
abstract |
(57) SUMMARY Substrates and assays for the identification of activators and inhibitors of caspases are provided. The substrate is a fusion protein containing two green fluorescent proteins (GFP) with a linker peptide containing at least one caspase cleavage site. The complete fusion protein shows fluorescence resonance energy transfer (FRET) between GFP. When the linker peptide is degraded by caspase, two GFPs are separated, and FRET is attenuated. |
