PROBLEM TO BE SOLVED: To provide an efficient method for inserting a DNA (LPS) serving as a scaffold for introducing a large-sized DNA into a genomic vector of a bacterium of the genus Bacillus or a thermophile. To do. SOLUTION: When LPS is inserted into a host genome by homologous recombination, a homologous sequence used for recombination is replaced with LPS. By using a marker gene sequence that is not S, It can be easily performed using a vector in which only one LPS fragment is inserted into a vector used for recombination. |
