PROBLEM TO BE SOLVED: To provide an improved humanized immunoglobulin which is substantially non-immunogenic in humans and which is easily and economically produced in a form suitable for therapeutic preparations and other uses. And a therapeutic pharmaceutical composition using the same. SOLUTION: A framework region capable of binding an antigen with an affinity of at least 10 8 M −1 and derived from human acceptor immunoglobulin light and heavy chains, and from donor globulin Kabat et al. (“Sequences of Proteins”). of Immuno logical Interest ", Kabat, E. et al., USDepartment of H ealth and Human Services, (1983)) and a method for producing a humanized immunoglobulin having a complementarity determining region (CDR). The method includes replacing at least one non-CDR framework amino acid of an acceptor immunoglobulin with a corresponding amino acid from a donor immunoglobulin, wherein the substitution is made at a predetermined position. |
