titleOfInvention Chromosome dynamic analysis method
abstract (57) [Summary] [Problems] Conventionally, it was difficult to observe the state of chromosomes, particularly higher plants, for example, wheat, etc. A chromosome dynamic analysis method capable of observing the movement of a parent-derived chromosome in a tissue that maintains a favorable morphology of a hybrid. A sample derived from an individual hybrid plant to be analyzed is embedded with a sample embedding compound having a specific property, whereby an embedded sample is obtained. And a labeled probe obtained by labeling nucleic acids derived from at least two types of analytical species suspected of providing nucleic acids with different labeling substances, respectively, and allowing the labeled probe to penetrate the embedded sample. And maintaining the labeled probe under conditions suitable for specifically annealing a nucleic acid derived from a hybrid plant individual having a sequence complementary to the sequence of the probe, whereby the labeled probe and the hybrid plant individual are maintained. A hybrid with the derived nucleic acid is formed, and the dynamics of the chromosome in the hybrid plant individual is observed via a signal derived from the labeling substance of each labeled probe in the formed hybrid. Chromosome dynamics analysis method.
