abstract |
PROBLEM TO BE SOLVED: To provide a novel acid peroxidase having high activity under extremely low pH conditions such as exhibiting a maximum activity at pH 2.3, and a gene encoding such acid peroxidase. SOLUTION: A novel acid peroxidase (TAP) is isolated and purified by culturing a basidiomycete Thermitomyces albuminosas ATTC42010. Further, RACE-PCR is performed by extracting mRNA from the above strain and designing primers based on the N-terminal amino acid sequence information of purified TAP, thereby obtaining a full-length TAP gene. The TAP gene showed only very low homology (approximately 30%) to the existing basidiomycete-derived peroxidase, but to the peroxidase Dyp from the ascomycetous Geotricum candium which had the highest activity at pH 3.2. And TAP showed 56% homology in the amino acid sequence. |
