abstract | (57) [Summary]nThe present invention provides methods for discovering molecules that target an essential aspect of eukaryotic gene expression-the formation of mRNA 5 'cap m7GpppN. The basic principle of the present invention is to use different strains of the test organism which differ only in the composition or origin of the essential capping enzyme. In addition, the invention provides isogenic yeast strains wherein all of their capping activities are derived from pathogenic bacteria or mammals. These strains form the basis of a differential growth inhibition assay to identify molecules that specifically target the fungal capping mechanism. The invention also provides a method for in vitro screening for molecules that inhibit fungal RNA triphosphatase, an essential enzyme that catalyzes the first of three steps in cap synthesis. |
