abstract | (57) [Summary] The present invention relates to the synthesis of an extracellular protease of Acremonium chrysogenum having CPC acetylhydrolase activity and a deacetylated derivative of cephalosporin C and inactivation of a gene to increase the yield of cephalosporin Regarding its use in. This method involves the preparation of an enzyme with CPC-acetylhydrolase activity starting from the culture medium of the filamentous fungus A. chrysogenum C10 (ATCC 48272) (CPC-AH B) and the biochemical characterization of said enzyme. In addition, the cloning and sequencing of genomic DNA fragments and other complementary DNA (cDNA) containing the cahB gene encoding the enzyme are described. The production of CPC-AHB enzyme activity in E. coli by use of this gene in the cDNA form is also described. Finally, a method for inactivating the cahB gene to increase the yield of cephalosporin in A. chrysogenum is included. |
