titleOfInvention | Method for measuring ligand / target affinity promoted by centrifugation |
abstract | (57) Abstract Centrifugation is used to induce and / or enhance the binding of macromolecular targets to small molecule ligands or larger biomolecules (alone or as a mixture). Due to the enhanced binding, the method of the present invention allows for the detection of a ligand that binds to a target substance and enhances the design of the ligand. The method is based on centrifugal force that establishes a differential and selective concentration gradient between the macromolecular therapeutic target and the desired ligand. Once formed, information about the self-sorting binding event is derived by analyzing the differential gradients of macromolecules and ligands in situ, or by fractionating the gradient into individual samples for further analysis. I do. Various methods or combinations thereof can be used to find enhanced binding ligand levels. |
