titleOfInvention | A method for large-scale cDNA cloning and sequencing by repeating subtraction |
abstract | (57) [Summary] The present invention provides a cDNA sequencing method by repeating subtraction, comprising the following steps: (1) A step of preparing a high-quality cDNA original library using a tissue of an organism as a material (2) homogenizing the cDNA original library of step (1) according to the graded Cot value, thereby obtaining a cDNA homogenized library corresponding to the graded Cot value; (3) said step (4) using the sequenced cDNA clones to hybridize and subtract the homogenized cDNA library from the above step; Steps (3) and (4) are 1-50 0 times repeated step. Further, a preliminary subtraction hybridization step may be included in the method. By using this method, certain tissues or certain cDNAs can be efficiently and comprehensively sequenced. |
