titleOfInvention | Multi-domain polynucleotide molecule sensor |
abstract | (57) [Summary]nA multi-domain polynucleotide responsive to a signal agent is designed and constructed to have at least three domains, which may be partially or completely overlapping or non-overlapping. Yes: actuator (catalytic or reporter) domain, bridging domain and receptor domain. In an exemplary embodiment, a signaling agent, such as a chemical ligand, interacts with the receptor domain, which changes conformation or otherwise affects the cross-linking domain, such that the actuator The activity, catalytic or reporter function of the domain is stimulated or suppressed. In certain ribozyme embodiments, for example, the cross-linking of a ligand to a receptor domain functions to cause a conformational change within the cross-linking the pre-existing catalytic and receptor domains by a novel structural cross-link. The binding results in a ligand-specific molecular sensor composed of RNA, whose structural recognition is indicative of the activity of the adjacent ribozyme. Methods for allosterically selecting other multi-domain polynucleotides typically involve mixing and adapting the domains to optimize cross-linking or other signal response and / or reporter activity. . |
