abstract | (57) [Summary]nMethods for increasing the target-specific toxicity of an agent include pre-targeting the enzyme to a target site in a mammal, and a cytotoxic agent known to act at the target site, or one that is converted to the agent in situ. Wherein the drug is detoxified to form a less toxic intermediate using the normal metabolic processes of the mammal, whereby the detoxified intermediate is pre-targeted. It is converted back to a more toxic form by the given enzyme and has high cytotoxicity at the target site. In addition, an increase can be achieved by pre-targeting an enzyme that converts the prodrug into a cytotoxic drug at the target site. Kits for use in the methods of the invention are also provided. The methods and kits of the present invention can reduce the dose of cytotoxic agent, maximize activity at the target site, and minimize systemic side effects. |
