(57) Summary The stereochemical center of a polyketide can be altered by substitution of the ketosynthase domain in the polyketide synthase (PKS) enzyme that produces the polyketide. The specificity of the AT domain of PKS is determined by the hypervariable region, which can be substituted or altered, and the specificity of the AT domain is increased from a naturally occurring extension unit to another naturally or non-naturally occurring extension. Change to a unit. Non-naturally occurring extension units, including methylmalonyl N-acetylcysteamine thioester, can be incorporated into polyketides in recombinant host cells or cell-free systems to create polyketides. |
