A pharmaceutical composition in the form of a tablet or capsule provides a combination of temporary and controlled delivery of a drug to a patient for effective therapeutic results. The pharmaceutical composition includes a drug, a gas generating component, a swelling agent, a viscosity-imparting agent, and a gel-forming polymer. Swelling agents belong to the class of compounds known as superdisintegrants (eg, cross-linked polyvinyl pyrrolidone or cross-linked sodium carboxymethyl cellulose). Initially, the viscosity-imparting agent, and then the gel-forming polymer, forms a hydrated gel matrix that traps the gas, thereby suspending the tablet or capsule, so that it becomes the upper surface of the stomach or small intestine ( Spatial control) is stagnated. At the same time, the hydrated gel matrix created a meandering diffusion path for the drug, thereby resulting in sustained release (temporary control) of the drug. A preferred once-daily ciprofloxacin formulation is 69.9% ciprofloxacin base, 0.34% sodium alginate, 1.03% xanthan gum, 13.7% sodium bicarbonate, 12.1% cross-linked polyvinyl. Including pyrrolidone, and optionally other pharmaceutical excipients, the formulation is in the form of coated or uncoated tablets or capsules. |
