Methods for enhancing and limiting gene expression |
abstract |
(57) [Summary]nThe present invention provides a new method of gene therapy for a limited area such as a tumor. These methods comprise the steps of (a) placing the gene in a plasmid vector that is activated by a heat or light inducible promoter, and (b) including a tetracycline responsive fusion protein that functions as a transcriptional activator. Modifying the vector and (c) modifying the vector by including a DNA sequence that reduces or eliminates expression of the gene in normal peripheral cells. A series of vectors for persistent and controllable expression are also provided. New vectors for gene therapy of localized and metastatic breast, ovarian, and prostate cancers are also provided. |
