titleOfInvention Methods for detecting potential cytotoxicity of compounds
abstract (57) [Summary]nThe present invention relates to a test procedure for screening a test compound for potential cytotoxicity, comprising the following steps. (A) producing liposomes from one lipid or a mixture of lipids to simulate the cell membrane composition of interest, wherein the liposomes are in an aqueous medium having a pH that is the internal pH of the liposomes. To be manufactured. (B) mixing the liposome with an aqueous test medium having a pH at least about 0.5 pH units above the internal pH of the liposome, whereby a pH gradient exists between the test medium and the interior of the liposome. (C) In the step of incorporating the fluorescent dye into the test medium, the dye has a tendency to migrate to the liposome by a pH gradient, and the dye is pH sensitive such that the fluorescence of the dye is quenched inside the liposome. (D) In the step of admixing the test compound with the test medium, if the test compound disrupts the structure of the liposome or otherwise causes the dissolution of the pH gradient between the test medium and the interior of the liposome, the fluorescence of the dye Not quenched. (E) measuring the fluorescence of the test medium and comparing it with the fluorescence of the control.
