titleOfInvention | DNA sequences, molecules, vectors and vaccines for feline calicivirus disease and methods of making and using them |
abstract | (57) [Summary]nProvided is a nucleotide sequence obtained from the genome of the capsid gene of FCV2280 strain, wherein the splicing site has been modified to be deleted or inactivated. These nucleotide sequences include a DNA sequence encoding a polypeptide, such as the capsid gene of FCV2280 strain. Therefore, although their coding sequences can be transcribed in the nucleus of eukaryotes, the DNA coding sequences are not altered by the natural splicing machinery of the organism and the amino acid structure of the expressed protein is Not changed. These recombinant molecules can be incorporated into FHV-1 vectors used to infect cell cultures, among others, for the development of vaccines, especially vaccines for preventing or treating FCV disease. Further provided is a method of obtaining a nucleotide sequence encoding a polypeptide, which is originally transcribed in the cytoplasm of a cell, and in particular, a DNA sequence encoding an FCV capsid protein. In addition, these sequences have been modified to allow their transcription in the nucleus of a eukaryotic cell, and the modified coding sequence is not altered by the eukaryotic cell splicing machinery. |
