(57) [Summary]nThe present invention provides a genetic method for anchoring a polypeptide to the yeast cell wall in an accessible form for binding to macromolecules. The combination of fluorescence activated cell sorting and this method provides a means to select proteins with increased or decreased affinity for another molecule, altered specificity, or conditioned binding. Also provided is a method of genetically fusing the N-terminus of a polypeptide of interest to the C-terminus of a yeast Aga2p cell wall protein. Attachment of scFv antibody fragments to Aga2p agglutinin effectively mimics cell surface display of antibodies by B cells in the immune system for affinity maturation in vivo. T cell receptor variants that are efficiently displayed on the yeast cell surface are isolated by this method, which may provide a means to alter T cell receptor binding affinity and specificity by library screening. This selection method also identifies proteins with enhanced phenotypic characteristics (proteins displayed at higher levels, proteins secreted with higher efficiency, and more stable proteins). |
