abstract | (57) Abstract: A method for detecting a target nucleic acid molecule, comprising the following steps: (a) (i) a target nucleic acid molecule; (ii) a single-stranded nucleic acid probe containing a cleavable linkage; (iii) A) an enzyme capable of cleaving the probe portion of the double-stranded target-probe complex at the cleavable linkage; and (iv) a mixture comprising ribosomal proteins and / or spermine, wherein the target nucleic acid and the probe hybridize to each other. And reacting under conditions and for a time sufficient to allow formation of a double-stranded target-probe complex, followed by cleavage of the probe and cycling of the target to a new uncleaved probe. Releasing the one or more portions of the cleaved nucleic acid probe from the target-probe complex; and (b) the nucleic acid probe. Cut portions of the probe will determine whether it is produced, comprising thereby detecting the presence of the target nucleic acid, a method is provided. |
