titleOfInvention Transsomatics with gene transfer to breast epithelial cells
abstract (57) Abstract Methods of transferring valuable compounds, eg, genes encoding pharmaceuticals, to the mammary gland to create new compounds in milk or alter the composition of milk are described. In this method, a packaging cell line that produces virus-derived particles is injected into the mammary gland. The packaging cells will attach and survive in the mammary gland for a specific period of time. The cells survive and they will provide a continuous source of virus-derived particles and will transfect the largest number of mammary epithelial cells. After being present in the mammary gland for a certain period of time, its particles and packaging cells will be destroyed by natural mechanisms, and trans-infected mammary epithelial cells will alter the genes encoding valuable compounds or the composition of milk Keep expressing the gene. One or more genes, including DNA sequences that contribute to efficient production of the active compound or contribute to its stability, can be trans-infected. The packaging cells and virus-derived particles used in this method may be those that transfect divided or undivided cells and may be used alone or together. This method can be used alone or in combination with other novel methods designed to ensure that virus-derived particles are correctly located to trans-infect breast epithelial cells. Increasing trans-infection of mammary epithelial cells with viral particles will result in higher concentrations of valuable compounds in milk.
