PROBLEM TO BE SOLVED: To provide a recombinant DNA encoding a BstYI restriction endonuclease and a BstYI methyltransferase, and an E. coli containing the recombinant DNA. The present invention also relates to the expression of the endonuclease and the methylase in E. coli cells and a purification method thereof. The present invention relates to a BstYI methylase gene and a BstYI methylase gene. A method of cloning stYI restriction endonuclease and expressing it in E. coli, and selecting Bacillu by methylase selection and inverse PCR amplification of adjacent DNA. Provided is a method for cloning BstYI restriction endonuclease from S. stearothermophilus in E. coli. After methylase selection, DNA A methylase gene having high homology to amino-methyltransferase (N4-cytosine methylase) was found in the library and was named BstYI methylase gene (bstYIM). |
