abstract | (57) [Problem] To provide a method for cloning T cells of unknown specificity and a method for identifying a peptide ligand recognizing the same. SOLUTION: A T cell of unknown specificity is obtained by combining a T cell with a combinatorial random peptide library (Xn peptide library (n is the number of amino acids)) of peptides consisting of 19 kinds of natural amino acids except cysteine. A method for cloning T cells of unknown specificity, characterized by culturing with cells expressing the major histocompatibility complex (MHC) class II antigen derived from the same individual and in which DNA synthesis has been stopped, and T cell clones obtained therefrom The T cell clone is recognized by examining the growth stimulating activity using an Xn peptide library in the presence of interleukin and MHC class II-expressing cells in which DNA synthesis derived from the same individual as the T cell has been stopped.
