(57) [Summary] [Objective] Aconitine is conjugated with bovine serum albumin as a carrier protein by the carbodiimide method. This is used as an antigen to immunize mice. Post-immunization spleen cells are isolated and fused with myeloma cells using polyethylene glycol. Only fused cells (hybridomas) are selected in HAT medium. The hybridoma is cloned by the limiting dilution method. From these, selection is performed using an aconitine-ovalbumin (EA) complex, and a hybridoma producing an anti-aconitine monoclonal antibody is isolated. [Effect] This antibody has a very high specificity, so that it can be By using for SA, quantification with high reproducibility, high sensitivity, and no pretreatment is possible. |
