abstract | (57) [Summary]nC2 symmetric and asymmetric chemical libraries for use in homodimerization and heterodimerization of proteins and receptors are constructed by liquid phase methodology. A representative library is prepared by symmetrically coupling the assembled fragment and a mixture of tethered dicarboxylic acids in a 60-10 sublibrary format. At each step of the three-step reaction sequence, reactants, unreacted starting materials, reagents and by-products are removed by simple liquid-liquid or liquid-solid extraction, and the desired intermediates and final library are purified to high purity ( ≧ 90-100%) and in amounts of a few milligrams, independent of reaction yield and without careful reaction optimization. The synthesis of the second prototype library uses an olefin metathesis reaction, linking a linker tether and randomizing its length by combination. This approach offers the opportunity to rapidly perform statistically controlled mixtures of homo and hetero dimers with significant diversity. |
