titleOfInvention | Methods for producing proteins |
abstract | (57) [Summary]nThe present invention provides a method for producing a polypeptide as an inclusion body in a bacterial host cell. The method of the present invention E. coli thioredoxin or modified E. coli. This is accomplished by forming a genetic construct comprising the genetic sequence encoding the polypeptide operably linked to the genetic sequence of an inclusion partner protein such as E. coli thioredoxin, such that the host cell containing the genetic construct is Produces the polypeptide as an internal inclusion. The method of the present invention facilitates rapid isolation and purification of the recombinant protein. In addition, the methods of the invention are useful for polypeptides or proteins that are small and typically difficult to express, as well as E. coli. It may be useful for producing proteins that are toxic to host cells such as E. coli. The present invention also provides the plasmids, vectors, and host cells used in the present invention for the production of polypeptides, and methods of producing polypeptides using these vectors and host cells. The present invention further provides methods for producing protein molecular weight ladders for use in protein gel electrophoresis, as well as proteins and protein molecular weight ladders produced by these methods. |
