titleOfInvention | Internal positive control for nucleic acid amplification |
abstract | (57) [Summary]nThe present invention relates to an improved method for amplifying a target sequence of a target polynucleotide. The method comprises mixing a sample suspected of containing the target polynucleotide with a reagent for amplifying the target sequence and subjecting the mixture, if present, to conditions under which the target sequence is amplified. The improvement comprises adding to the mixture a control oligonucleotide and a control polynucleotide having a sequence capable of hybridizing to the control oligonucleotide. When the control oligonucleotide binds to the control polynucleotide, the ability of the primer to extend along the control polynucleotide is reduced. Optionally, the control oligonucleotide is part of a control polynucleotide. This method finds particular application in nucleic acid amplification and detection. |
