abstract | (57) [Summary]nThe present invention provides methods for use in identifying, analyzing, and typing polymorphic DNA fragments, particularly minisatellite, microsatellite, or STR DNA fragments. In particular, the present invention uses a DNA polymerase, more particularly a thermostable DNA polymerase, and most particularly a Thermotoga polymerase or a variant or derivative thereof, thereby producing a minisatellite, microsatellite, or STR DNA molecule. Provide methods that can be amplified and analyzed for polymorphisms. The present invention also relates to polymerases having reduced, substantially reduced, or eliminated ability to add non-templated 3 'nucleotides to a synthesized nucleic acid molecule. According to the present invention, such reduction or elimination may be achieved by modifying or mutating the desired polymerase. |
