abstract | (57) [Problem] To provide a method capable of detecting various mutations in a nucleobase at a predetermined position with high sensitivity and high accuracy. SOLUTION: A target nucleic acid and a primer are annealed. Next, a primer extension reaction is performed using a reagent containing a terminator nucleotide and a non-terminator nucleotide. The terminator nucleotide is complementary to the intended wild-type base. In the target nucleic acid, if the first base on the 3 ′ side that is not paired with the primer is wild-type, a terminator complementary thereto binds to the 3 ′ end of the primer, and the primer extension reaction stops. On the other hand, when the first base is mutated and not wild type, a non-terminator is incorporated into the primer, and the primer extension reaction proceeds. By analyzing the product after the primer extension reaction, the presence or mutation of the wild-type base can be known. |
