abstract |
PROBLEM TO BE SOLVED: To immobilize a DNA fragment prepared in advance separately on a solid-phase carrier surface by a rapid reaction, and a reaction product can stably maintain a bond. Develop a method to obtain a DNA chip that does not require a blocking step. SOLUTION: A solid support having a surface into which a boron-containing compound or a divalent partial structure that forms a cyclic bond by reacting with the boron compound is introduced, and a cyclic compound that reacts with the boron compound at one end thereof. Contact between a divalent partial structure forming a bond or a DNA fragment into which a boron compound has been introduced in a liquid phase, thereby forming a bond by a reaction between the boron-containing compound and the divalent partial structure. A method for immobilizing a DNA fragment on a solid-phase carrier surface, a DNA chip obtained by this method, and a method for detecting a nucleic acid fragment having complementarity to a DNA fragment on a DNA chip using the DNA chip . |
