abstract | PROBLEM TO BE SOLVED: To specifically detect a specific DNA fragment of interest from a mixture of various types of DNA fragments, and to easily detect the presence or absence of a target DNA sequence, the presence or absence of a mutation, or the type of mutation. SOLUTION: After a sample DNA fragment is denatured into a single strand, the sample DNA fragment is allowed to bind to a target DNA fragment in a column having a stationary phase on which an immobilized ligand is fixed. By contacting, only the target DNA fragment is specifically bound to the ligand, the other DNA fragments are washed away, and then the binding force between the ligand and the DNA fragment is reduced by gradually increasing the column temperature.
