abstract (57) [Summary] [Object] To produce trans-4-hydroxy-L-proline useful as a pharmaceutical raw material or a food additive in an industrially advantageous manner. Also, it enables specific and sensitive automatic separation and analysis of trace amounts of optical isomers of prolines. The present invention relates to L-proline in an aqueous medium in the presence of an enzyme source that catalyzes a hydroxylation reaction of L-proline to trans-4-hydroxy-L-proline (t4HYP), ferrous ion and diketoglutaric acid. In a method for producing t4HYP, a method for analyzing L-proline 4-position hydroxylase useful for the production method, and a method for analyzing prolines using HPLC, characterized in that the eluate is converted to t4HYP, the eluate is supplied in a liquid sending line. Buffer solution containing a metal chelating agent, and reacting prolines in the eluate with 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole (NBD) chloride to produce NBD-proline And a method for detecting and quantifying prolines.
