Production of naturally folded secreted proteins by co-secretion of molecular chaperones |
abstract |
PROBLEM TO BE SOLVED: To provide a water-soluble spontaneous folding after expression in prokaryotic cells, which can be easily carried out and does not require laborious in vitro post-treatments such as lysis, reduction and reconstitution of inclusion bodies. An object of the present invention is to provide a method for producing a selected eukaryotic polypeptide. SOLUTION: a) a step of culturing a prokaryotic cell containing an expression vector encoding a polypeptide containing a prokaryotic signal sequence at the N-terminus; b) a step of secreting the polypeptide into the periplasm or medium; c) cleavage of the signal sequence And The step of isolating the polypeptide from the periplasm or the medium provides a method for producing a naturally folded eukaryotic polypeptide comprising two or several cysteines linked by disulfide bridges. |
