titleOfInvention | Method for obtaining cells for measuring transcriptional regulation |
abstract | (57) [Problem] To provide a cell or the like which can be used to evaluate the effect of a chemical substance on the transcription regulation ability of a ligand-responsive transcription regulator. (1) An animal cell into which a DNA containing a gene encoding a ligand-responsive transcriptional regulator is introduced simultaneously with or before or after this step (1), or A step of introducing a DNA containing the following gene (a) and the gene (b) on the same molecule into an animal cell capable of expressing a gene encoding a responsive transcription regulator; A reporter gene operably connected downstream of a transcription regulatory region consisting essentially of a transcription regulatory factor recognition sequence and a minimal promoter operable in animal cells. (B) A cell selection marker gene that can function in animal cells. (2) measuring the transcriptional regulation ability, which comprises a step of recovering, from the transformed cells obtained in the step (1), cells in which the introduced DNA is stably retained in the cells. How to obtain cells for use, etc. |
