(57) [Summary]nDNA vaccines that incorporate a gene sequence encoding a sorting signal that directs the expressed antigen to specific organelles facilitate the addition of the antigen to Class I or Class II MHC molecules for immune presentation. These vaccines are nucleic acid constructs of a gene sequence encoding a protein or peptide antigen and a classification signal that directs the expressed antigen to the ER or endosomal-lysosomal compartment in the cell. The resulting construct can be used as a naked DNA vaccine packaged in liposomes or coated on colloidal gold particles. The construct is also delivered in an expression vector that is expressed in cells of the organism to be immunized. |
