abstract | (57) [Summary]nThe present invention relates to a method for substantially improving the stereoselective synthesis of nucleoside analogs of β-anomers. In the method according to the invention, the introduction of the phenylseleno group into the blocked lactone sugar precursor is selected such that the desired phenylseleno group is introduced on the blocked lactone side away from the blocking group. Stereospecific introduction of a phenylseleno group into a sugar precursor leads to the formation of nucleoside analogs, especially 2 ', 3'-dideoxy and 2', 3'-dideoxy-2 ', 3'-didehydronucleoside analogs. The synthesis can be performed in an extremely high yield. In certain preferred embodiments, the lactone blocked with the preferred phenylseleno groups is obtained in an amount of approximately 90% or more of the total stereoisomer produced. In a more preferred embodiment, the preferred amount of stereoisomers is at least about 95%, more preferably at least about 97%, of the total amount of phenylseleno-blocked lactone produced. |
