titleOfInvention Use of Gram-positive bacteria to express recombinant proteins
abstract (57) [Summary] A new system for cloning and expression of genes in Gram-positive bacteria. The expression system is based on the finding that many Gram-positive bacteria sort proteins on their cell surface by an N-terminal signal sequence acting in cis and a C-terminal anchor region. In particular, the cell sorting signal of streptococcal M6 protein, a well-known surface molecule, is used to create a gram-positive bacterial expression system named SPEX (Streptococcal Protein Explession). Expression is achieved by cloning the gene of interest into an appropriate SPEX cassette and then stably introducing it into a bacterial host such as S. gordonii which is commensal for humans. Depending on the SPEX vector used, the recombinant protein is either fixed to the cell wall before being dissociated by specific internal proteolytic cleavage, or is secreted into the culture medium during bacterial growth. The secreted protein should be protected from proteolysis by using host bacteria without extracellular proteases. In this system, several expression vectors also produce specifically tagged recombinant proteins, which allow for the purification of the product in a single step.
