abstract | (57) [Summary]nDisclosed are methods for forming a conjugate, preferably a protein / polysaccharide conjugate. In this method, a reagent for providing a functional group on a polysaccharide molecule is reacted with the polysaccharide. After the functional groups are in place, the polysaccharide is reacted with homobifunctional and heterobifunctional vinyl sulfones to form vinyl sulfone derivatized polysaccharides. The vinylsulfone-derivatized polysaccharide is then reacted with the protein to form a conjugate. If desired, the functional group may be used to form a protein derivative prior to the conjugate formation reaction step. In another embodiment, the protein is functionalized with a reactive group and then derivatized with a vinyl sulfone group to form a vinyl sulfone derivatized protein. Next, the protein is reacted with the polysaccharide to form a conjugate. Optionally, the polysaccharide may be derivatized with a reactive group prior to the conjugate formation reaction. In another method, the polysaccharide is directly derivatized using the cross-linking agent pendant vinyl sulfone. Next, the vinyl sulfone-derivatized polysaccharide is coupled to a protein, which may or may not be functionalized. Similarly, protein derivatives may be formed directly using the cross-linking agent pendant vinyl sulfone, and then attached to a polysaccharide molecule, which may or may not be functionalized. . |
