abstract | (57) [Summary]nThe present invention relates to variants of the jellyfish Aequorea Victoria Green Fluorescent Protein (GFP) which have a specific green fluorescence at least 5-fold, preferably more than 20-fold, of the wild-type protein. In another embodiment, the invention comprises a mutant blue fluorescent protein (BFP) that emits increased blue fluorescence. The invention also includes the expression of nucleic acids encoding the mutant GFP or BFP in a wide variety of engineered host cells, as well as the isolation of engineered proteins with increased fluorescent activity. The novel mutants of the present invention allow for significantly more sensitive detection of fluorescence in engineered host cells than is possible with GFP or known variants thereof. Thereby, the mutant fluorescent protein provided herein is a cell and tissue specific of a GFP or BFP mutant or a chimeric protein comprising a GFP or BFP mutant fused to a regulatory sequence or to a second protein sequence.
